The metabolic pattern of utilization of [1,2,3,4-14C, methyl-3H] -butyrobetaine and d-and l-[1-14C, methyl-3H]carnitine has been examined with variously grown resting cell suspensions of Acinetobacter calcoaceticus and Pseudomonas putida. Ps. putida grown on d, l-carnitine as the sole source of carbon, degraded only l-carnitine with stoichiometric accumulation of glycinebetaine. Alternatively, when grown on -butyrobetaine, Ps. putida rapidly metabolized -butyrobetaine, and to a lesser but significant extent, both d-and l-carnitine with equivalent formation of trimethylamine and degradation of the betaine carbon skeleton. Ac. calcoaceticus grown on either d,l-carnitine or -butyrobetaine, effectively utilized all three betaines at nearly the same rates. Disappearance of each of these quarternary ammonium compounds was accompanied by stoichiometric formation of trimethylamine and degradation of the carbon backbone. Utilization of the betaines and corresponding formation of trimethylamine by resting cell suspensions of appropriately grown Ac. calcoaceticus and Ps. putida, was essentially abolished under conditions of anaerobiosis and severely impaired in the presence of sodium cyanide, sodium azide, 2,4-dinitrophenol or 2,2-bipyridine. The results of the present investigations with resting cell suspensions of both Ac. calcoaceticus and Ps. putida do not support an earlier suggestion that -butyrobetaine degradation in these organisms proceeds by its prior hydroxylation to l-carnitine. Indeed, disrupted cell-free preparations of Ac. calcoaceticus and Ps. putida grown on either d,l-carnitine or -butyrobetaine showed no detectable -butyrobetaine hydroxylase activity. 相似文献

Summary The inducibility of cytochrome P-450 in Acinetobacter calcoaceticus by some compounds known as typical inducers of hepatic cytochromes P-450 was investigated. Besides biphenyl also indene and phenanthrene are inducers, whereas compounds of the so-called phenobarbital type are not. Biphenyl appears to be the most effective inducer with regard to the yield of cytochrome P-450/mg of cell protein. By addition of the compounds in the vapour phase an induction of the protein by naphthalene could be demonstrated. The results are indicative of the existence of bacterial cytochromes P-450 that resemble hepatic cytochromes. 相似文献

1. Harvester ants are major seed predators in arid environments. However, given that many harvester ants are partly omnivorous and therefore potentially attracted to the elaiosomes of myrmecochorous seeds, it is unclear if these ants act as predators or dispersers when removing myrmecochorous seeds. 2. We describe the outcomes of interactions between the harvester ant, Pogonomyrmex naegelii, and myrmecochorous plant, Microstachys serrulata, in a Brazilian savanna. We: (i) evaluated the role of elaiosome in seed removal by P. naegelii; (ii) investigated the fate and viability of removed seeds; (iii) tested if soils associated with P. naegelii nests are nutrient-enriched; (iv) compared seedling survival; and (v) the density of seedling and adult M. serrulata nearby to P. naegelii nests compared with those away from these nests (i.e. controls). 3. Rates of removal of M. serrulata seeds were two-fold higher with elaiosomes than without. The ant attractant oleic acid was the dominant fatty acid in elaiosomes, but it was absent from seeds. Removed seeds are taken into nests, and Tetrazolium tests indicated that 95% of seeds remain viable. Soils associated with P. naegelii nests were not nutrient-enriched, and seedling survival was similar nearby to P. naegelii nests compared with control areas. However, densities of both seedling and adult M. serrulata were higher nearby to P. naegelii nests than in control areas. 4. Our findings show that P. naegelii switches its role from seed predator for most plant species to be the dominant seed disperser for M. serrulata, playing a key role in the distribution of adult plants. 相似文献

Oxysterols comprise a very heterogeneous group derived from cholesterol through enzymatic and non-enzymatic oxidation. Among them, 7-ketocholesterol (7-KC) is one of the most important. It has potent effects in cell death processes, including cytoxicity and apoptosis induction. Mesenchymal stem cells (MSCs) are multipotent cells characterized by self-renewal and cellular differentiation capabilities. Very little is known about the effects of oxysterols in MSCs. Here, we describe the short-term cytotoxic effect of 7-ketocholesterol on MSCs derived from human adipose tissue. MSCs were isolated from adipose tissue obtained from two young, healthy women. After 24 h incubation with 7-KC, mitochondrial hyperpolarization was observed, followed by a slight increase in the level of apoptosis and changes in actin organization. Finally, the IC50 of 7-KC was higher in these cells than has been observed or described in other normal or cancer cell lines. 相似文献

The occurrence of herbivores in nature is limited by biotic and abiotic factors which affect their development and survival. Udranomia spitzi is an endemic butterfly in the Brazilian savanna (cerrado) that feeds on young leaves of two sympatric plants, Ouratea hexasperma and O. spectabilis. It is not known which factors affect the occurrence of larvae on their hosts. Therefore, in this study we: (i) evaluated the oviposition preference of U. spitzi; (ii) evaluated the larval performance in both Ouratea species; (iii) investigated plant phenology; (iv) investigated climate (temperature and rainfall); and (v) investigated plant architecture (measured as plant height) on the abundance of skippers. Results showed that U. spitzi immatures (eggs and larvae) were far more abundant (n = 41, 96.7%) on O. spectabilis, whereas on O. hexasperma, the number of larvae was negligible (n = 1). In the laboratory, U. spitzi performed better on O. spectabilis than on O. hexasperma. The occurrence of larvae was not related to host phenology or environmental variations, but rather to plant height, since 92.7% (n = 38) of larvae were found on small O. spectabilis trees. A previous study showed that U. spitzi was not influenced by biotic factors (aggressive ants) and this study showed that plant structure plays a major role in skipper choice. The CD95 (Apo-1/Fas)/CD95 ligand (CD95L) system is best characterized as a trigger of apoptosis. Nevertheless, despite broad expression of CD95L and CD95 in the developing brain, absence of functional CD95 (lpr mice) or CD95L (gld mice) does not alter neuronal numbers. Here, we report that in embryonic hippocampal and cortical neurons in vivo and in vitro CD95L does not induce apoptosis. Triggering of CD95 in cultured immature neurons substantially increases neurite branches by promoting their formation. The branching increase occurs in a caspase-independent and death domain-dependent manner and is paralleled by an increase in the nonphosphorylated form of Tau. Most importantly, lpr and gld mutants exhibit a reduced number of dendritic branches in vivo at the time when synapse formation takes place. These data reveal a novel function for the CD95 system and add to the picture of guidance molecules in the developing brain. 相似文献
